چکیده

Background and aims: Mendelian randomization studies have shown that triglyceride (TG)- lowering lipoprotein lipase (LPL) alleles low-density lipoprotein-cholesterol (LDL-C)-lowering independent beneficial associations on cardiovascular disease (CVD) risk. We aimed to provide further insight into this observation by applying analyses of genetically-influenced TG LDL-C levels plasma metabolomic profiles Methods: quantified over 100 measures in the Netherlands Epidemiology Obesity (NEO) study (N = 4838) Oxford Biobank (OBB) 6999) nuclear magnetic resonance (NMR) spectroscopy. Weighted genetic scores for via five LPL 19 were calculated dichotomized median, resulting four genotype combinations high/low LDL-C. performed linear regression using a two & times; design with group high as reference. Results: Compared individual groups lower or only, combined showed an overall additive pattern changes measures. Over different (p < 1.35 10-3) compared reference, effect sizes directionality being similar NEO OBB. Most notably, all very-low density (VLDL) LDL sub-particles lower. Conclusions: Our findings evidence TG-lowering top LDL-C-lowering has effects profile which is accordance reported CVD risk reduction.

Triglyceride-Increasing Alleles Associated with Protection against Type-2 Diabetes

Elevated plasma triglyceride (TG) levels are an established risk factor for type-2 diabetes (T2D). However, recent studies have hinted at the possibility that genetic risk for TG may paradoxically protect against T2D. In this study, we examined the association of genetic risk for TG with incident T2D, and the interaction of baseline TG with TG genetic risk on incident T2D in 13,247 European-Ame...

Distinctive audiometric profile associated with DFNB21 alleles of TECTA.

Genetic factors are thought to account for approximately one half of cases of childhood hearing loss, the majority of which is non-syndromic and not associated with other abnormalities. Seventy-seven percent of hereditary, non-syndromic, prelingual deafness is autosomal recessive, 22% is autosomal dominant, and 1% is transmitted as a matrilineal or X linked trait. So far, more than 30 distinct ...

Carvedilol: A Promising Drug Combined with Lipid-lowering Medications for Patients with Hypertension and Heart Failure

Background: Statins frequently cause myopathy especially in combination with fibrates, and physical activity is considered a trigger for the muscle disorder. Elevated plasma levels of creatine kinase (CK), lactate dehydrogenase (LDH) and aldolase, are the main indicators of the severity of myopathy. Carvedilol is commonly used with lipid-lowering drugs in the management of heart failure, hypert...

Common Kibra alleles are associated with human memory performance.

Human memory is a polygenic trait. We performed a genome-wide screen to identify memory-related gene variants. A genomic locus encoding the brain protein KIBRA was significantly associated with memory performance in three independent, cognitively normal cohorts from Switzerland and the United States. Gene expression studies showed that KIBRA was expressed in memory-related brain structures. Fun...
